Exposé of the Safety and Management of China's Biology and Virology Laboratory

So, what we are most concerned about is whether there was an accidental leak of the new coronavirus from the P4 lab's safety protection system in Wuhan? Before going into the details, let's start with the hardware, i.e. the main facility and equipment of P4 Lab, and gradually analyze the management system of P3 and P4 Labs in China.

This company's 2016 annual revenue reached 600 million yuan (including 55 million yuan in Taiwan), which means that this company's sales in the mainland region was 545 million yuan when the Wuhan P4 laboratory was built in 2016. It can be concluded that the equipment is so expensive and the technology involved is so complicated. Thinking of the quickly built Vulcan Mountain Hospital, Thur Mountain Hospital and the square cabin hospitals, I can only laugh.

Some of the information published on the Internet by this company gives us a glimpse into the true nature of P4 Labs' security measures.

Biosafety isolation systems in laboratories can be simply broken down into air systems and biosafety systems. In simple terms, researchers are required to conduct experiments on dangerous organisms, both to be able to conduct experiments (live, breathe, use equipment for research activities) and to protect against dangerous organisms. These protections include accidental leakage of dangerous organisms, harm to the researchers caused by accidental escape (animals), and harm to society caused by escape from the laboratory. So, the core equipment is the biosafety cabinet (including the high efficiency filter) and the air system. The technologies to design and manufacture these two core equipment are currently not available in China, they are in the hands of American companies.

Let's first take a look at the core equipment: the biosafety cabinet. There is a diagram of the equipment supplied to the Kunming Institute of Zoology.

Biosafety cabinet (also known as biological safety cabinet, SBC) is an important equipment commonly used in biological laboratories. It is widely used in medical, disease control, blood collection and supply, food hygiene, biopharmaceutical, environment monitoring and various biological experiment laboratories. High efficiency particulate (HEPA) filter inside the biosafty cabinet is used to filter exhaust gas and direct air flow downwards to avoid contamination of environment and laboratory operators by the infective organism, or the cross contamination between laboratory experiment materials.

HEPA filter functions as the key component in the biosafety cabinet by filtering bacteria and dust particulate from the gas pumped into and out of working area. The filter in the biosafety cabinet has to maintain high enough efficiency to protect human from biohazards. Currently, commonly used HEPA filters in bio-cabinets have 99.99% efficiency of capturing 0.3um dust particulates and ULPA (Ultra-Low Particulate Air) filters have efficiency of 99.9998%.

In term of integrity testing, according to AAF, MPPS (Most Penetrating Particle Size) leak test has to be carried out for each HEPA filter by passing through AstroScan station before leaving manufacturing facility. EN1822:2009 standard is strictly followed to automatically scan and categorize the class of the filters according to MPPS leak rate and overall efficiency for HEPA filters with different specifications and efficiency classes.

We provide unique identity for each HEPA and ULPA filter under MPPS test. Detailed test results, visualization of 3D test reports is transparent and reassuring to customer.

From the above statement, we can see the importance of biosafety cabinets for P4 and P3 laboratories, and the safe protection methods for viruses. The design of biosafety cabinets for bacteria, viruses, mammals (monkeys), experimental animals (mice) and livestock is only different in size, with basically same structure and principle.

In short, the laboratory must be a negative-pressure enclosed environment with an extremely strong enclosure (resistant to general destructive attacks, or higher-level attacks for military purpose. Of course, this is in the area of architectural design). After being sterilized layer after layer, laboratory personnel can enter the room and conduct scientific research in tight protective equipment (protective clothing, gloves, etc.).

Air environment inside the laboratory must be sterile and with constant temperature and humidity, with air outlet equipped with sterilization system. A negative pressure environment has to be maintained by negative pressure system. The experimental organisms or viruses must be kept inside of the biosafety cabinet (strictly prohibited to be removed from biosafety cabinet, if happens, alarm would be triggered). Then, could leakage accident like the one in movie Resident Evil happen?

Let's analyze: If it is true that the new coronavirus has spread to the seafood market due to a deliberate or unintentional leak in the course of the experiment, then several conditions are necessary: A, this person has access to the virus; B, this person can take the virus out of the P4 laboratory. So if A is established and this person has access to the virus, he/she must enter the P4 lab in a protective suit, then reach into the biosafety cabinet and get a live sample of the virus, then take it out of the biosafety cabinet, which would trigger a level 1 protection alert. Even if we believe that he/she has access to the virus and successfully removed it from the biosafety cabinet, can he/she take the sample out of the laboratory under multi-layer security measures and leave the P4 laboratory building? Obviously, the plot of the movie where the broken glass test tube causes the virus to leak is exaggerated and fictional.

The reality is that there have been hundreds of virus spills in the United States over the past few years, however, none of leakages have been able to get out of the labs and cause harm to society. According to a survey of more than 5,000 biosafety laboratories around the world with a total of 3,921 infections, 80% of laboratory infections were related to careless exposure of laboratory personnel to infectious aerosols, whereas only 20% were related to equipment accidents caused by lack of adequate level of safety protection systems or prevention control equipment. It can be said that the probability of accidental leakage of the new coronavirus is 0 after the installation of such a level of protection equipment, like the AAF biosafety prevention and control system, in the Wuhan P4 laboratory.

In accordance with the "Management Methods for the Construction and Operation of State Key Laboratories" and the "Rules for the Evaluation of State Key Laboratories", in 2016, the Ministry of Science and Technology commissioned the China National Center for Biotechnology Development to conduct an independent evaluation of a total of 75 state key laboratories in the field of biology and medicine, together with the China union of life science society. According to the assessment, after being reviewed in ministerial council meeting of Ministry of Science and Technology, the results of the assessment are now released.

I. Five-year overall development of national key laboratories in biology and medicine

According to the evaluation, through the five years of development, the overall level of the national key laboratories in the field of biology and medicine has risen rapidly and has become leading teams with some outstanding achievements and growing influence in various disciplines. The laboratories have pooled the best teams and advantageous resources in the country, and each laboratory carries out basic research around its own positioning and five-year goals, targeting disciplinary development, scientific frontiers and major national strategic needs.

The laboratory has become the backbone for major national tasks in this area. During the current assessment period, 75 laboratories have received a total of 20.8 billion yuan in funding, of which 15.5 billion yuan, or 74 per cent, of the total funding for national science and technology tasks, has reached an average of more than 278 million yuan for each laboratory. The scientific conditions and infrastructure of the laboratories have been greatly improved.

The laboratory has gathered, attracted and cultivated a group of outstanding scientific and technological talents and created a group of leading talents at the forefront of science. During the evaluation period, among the 75 laboratories, 15 new members were inducted to the Chinese Academy of Sciences, accounting for half of the new members of the Department of Life Sciences and Medical Sciences; 14 new members were inducted to the Chinese Academy of Engineering. Newly added innovative research groups and innovation teams reached 64 in key areas of the National Natural Science Foundation of China; 193 new chief scientists have been added to the National Key Basic Research and Development Program (973 Program), the National High Technology Research and Development Program (863 Program) and major scientific research programs; 274 new researchers have been added to the "Thousand People Program" (including the "Youth Thousand People Program"). 148 new recipients were funded by the National Outstanding Youth Science Fund, representing about 60 per cent of the total number of recipients of the National Outstanding Youth Science Fund during the same period.

The laboratories have achieved a large number of scientific research results and become an important source of original innovation. 75 laboratories have won 33 second place national natural science prizes, 1 outstanding, 12 first place and 65 second place national science and technology progress prize, 18 second place national technology invention prizes. In terms of academic impact, more than half of the evaluation experts considered more than 80 per cent of the participating laboratories to be at the leading or comparative level internationally.

II. Findings of the assessment

Twenty laboratories, including the State Key Laboratory for the Diagnosis and Treatment of Infectious Diseases, are classified as excellent State Key Laboratories.

Forty-six laboratories, including the State Key Laboratory of Virology, are classified as good State Key Laboratories.

The State Key Laboratory of Oncogenes and Related Genes, the State Key Laboratory of Silkworm Genome Biology, the State Key Laboratory of Forest Genetic Breeding, the State Key Laboratory of Cognitive Neuroscience and Learning, the State Key Laboratory of Experimental Hematology, the State Key Laboratory of Microbial Technology, the State Key Laboratory of Microbial Resources, the State Key Laboratory of Mycology, and other eight laboratories are undergoing rectification for a period of two years, during which the special fund for the State Key Laboratory will be suspended.

The State Key Laboratory of Medical Genetics has not passed the evaluation. According to the relevant provisions of the "Measures for the Construction and Operation of State Key Laboratories" and the "Rules for the Evaluation of State Key Laboratories", it is no longer included in the State Key Laboratories.

It is hoped that the participating laboratories, their supporting organizations and competent departments will take this assessment as a new starting point, seriously summarize the lessons learned, implement problem-solving methods and measures in response to the problems and recommendations pointed out by the assessment expert team, continuously improve the management level, take full advantage of the functions of key national laboratories in the area of gathering outstanding talents, undertaking major tasks and promoting collaborative innovation, and make greater contributions to the implementation of the innovation-driven development strategy and the building of a worldwide science and technology superpower.

This notice reveals a lot of critical information, let's decipher and dig into it.

1. The total amount of funding spent by the 75 laboratories amounted to 20.8 billion yuan, of which 15.5 billion yuan was spent on national science and technology projects, accounting for 74 per cent of the total amount of funding spent by laboratories, with an average of over 278 million yuan. The other $5.3 billion is of unknown origin and certainly not important. It is important to note that these laboratories are fully allocated by the national authorities in the name of national science and technology missions, with an average funding of more than $278 million per laboratory in 2016.

It is these laboratories that are really the mainstay of research in the biomedical field. First of all, let's remember these national laboratories associated with this new coronavirus induced outbreak among these mainstays, as follows.

"Management Measures for the Construction and Operation of State Key Laboratories" recently released

In order to further standardize and strengthen the construction and operation management of State key laboratories, on August 29, 2008, the Ministry of Science and Technology and the Ministry of Finance jointly issues the "Management Measures for the Construction and Operation of State Key Laboratories".

The State Key Laboratory Program has been organized and implemented since 1984, and has experienced rapid development in recent years with 220 State Key Laboratories, more than 10,000 permanent staff and a total value of more than 8 billion yuan of instruments and equipment. Promoted by such innovative systems as coordinated deployment, moderate new construction, regular evaluation, merit-based support and dynamic management, the State Key Laboratory has become an important base for our country's organizations to conduct high-level fundamental and cutting-edge technology research, recruit and train outstanding scientists and carry out academic exchanges, and has made outstanding contributions to exploring the frontiers of science and solving problems of great national interest. The original "Interim Measures for the Construction and Management of State Key Laboratories (2002)" has been executed for six years and has been widely recognized by the scientific and technological community, playing an important role in regulating the construction, operation and management of State key laboratories.

"The Outline of the National Medium to Long-term Science and Technology Development Plan (2006-2020)" clearly states that "the construction of key national laboratories should be strengthened and their overall level of operation and management should be continuously improved." Since 2008, in order to implement the spirit of the 17th National Congress and "The Outline of ..." and further strengthen the construction of key national laboratories, the state has set up special fund for national (key) laboratories to increase the steady support of key national laboratories from the three aspects: open operation, independent research on self-selected topics and the renewal of scientific research instruments and equipment. The establishment of special fund is helpful to create a research environment that tolerates failure, discards impatience and concentrates on research, is an important guarantee of good and rapid development of the State Key Laboratory, indicating that the State Key Laboratory has entered a new stage of development. In order to adapt to the new situation, the Ministry of Science and Technology and the Ministry of Finance have revised the original management methods on the basis of the experiences of the organizing and implementation of the National Key Laboratory Program, and have consulted the relevant departments and representatives of the National Key Laboratories on many occasions The revision process has taken nearly a year.

The new management measures also emphasize equipment sharing, data sharing, popularization of science, transformation of results, and cooperation with enterprises for state key laboratories.

The Ministry of Science and Technology and the Ministry of Finance will also jointly develop and issue the “Measures for the Management of Special Funds for State Key Laboratories”, in order to regulate the management of special funds and improve the efficiency of their use.

The "Measures for the Construction and Operation Management of State Key Laboratories" and the "Measures for the Management of Special Funds for State Key Laboratories" are public information and not listed here. We focus on the interpretation of the main amendments to the "Measures for the Construction and Operation Management of State Key Laboratories".

A. Special funds from the central government; procedures, requirements and methods for independent research topics, visiting scholar system, renewal of scientific instruments and equipment, and independent development.

B. The construction, operation and development of the laboratories should follow the principles of stable support, dynamic adjustment and regular evaluation, corresponding measures should be proposed.

C. Annual plans and summaries, random inspections of selected laboratories and the establishment of an "early warning mechanism" for operational management of the laboratory.

D. Reinforcement of the requirements for academic councils to ensure their functioning.

E. Strengthening of the responsibilities of the supporting institution by making sure that the supporting institution should give priority to development support to the laboratory.

From this, we can see that the key management departments of the laboratories are the daily management department, the academic committee and the supporting institution. Simply put, the daily management department is to manage the equipment and laboratory activities, the supporting institution is to manage the personnel and finance, and the academic committee is to manage the research direction and results evaluation. Based on this, all national laboratories in the list have a day-to-day management team (including the head of the laboratory, research team leads, etc.), an academic committee (which defines the research project and evaluates the results, and determines the publication of the results, such as the publication of papers, etc.), and a laboratory supporting institution. For example, the supporting institute of the National Key Laboratory of Virology is Wuhan University and Wuhan Institute of Virology. Finally, of course, there is the annual evaluation of the laboratories by the national organization. If the evaluation results do not meet the standards, funding for the following year will be affected.

For now, we do not argue whether virus leak could happen under this strict management and organization, but we can clearly see its impact and purpose: the scientific conduct of the laboratory staff is strictly controlled financially, organizationally and academically. All scientific activities are conducted collectively in the laboratory, and each group leader or researcher does not have the right to independently conduct scientific activities and publish scientific results including papers. The final published paper or the accolades received must have been earned as representatives (and of course actual contributions made) on behalf of the collective, the team. It was the organization that gave the honor, the organization that gave the award!

This strictly military-like laboratory R&D management system is also seen in technology companies such as Huawei and ZTE, because like these laboratories, they are just a government agency of the Chinese Communist Party and, in fact, an extremely important agency. In such an agency, not to mention the confidentiality regulations that bind every researcher, people are managed well by the management of supporting institution as the competent authority. Of course, most of these researchers wouldn't know what their research was for, what they did, would not know that it would become a weapon to kill themselves and their families in the future, would not know that they would be the next Zhao Yongfang at any moment.

Academian Tian Bo visited Wuhan Institute of Virology on July 5, 2011. This article tells the truth about exactly what this lab is.
