The call (literally)- to speak in a loud telling you to do something

"And now the call came to Buck in unmistakable accents. He, too, sat down and howled." This means that his instincts are taking over him and he is not civilized anymore.

"The wolf started on toward the place from where the call surely came, then returned to him, sniffing noses and making actions as though to encourage him." This means that the wolf followed his instincts or the call and relied on it over everything else.

The Call symolizes Buck's instincts and ancestral memories making him remember where he is supposed to be. The Call is telling Buck that his real home is the Wild, like the title says.
